Genome-scale esiRNA libraries

Our genome-scale esiRNA libraries are state-of-the-art RNAi reagents especially suitable for large-scale loss-of-function screening. Genome-scale collections of individual esiRNAs for human and mouse species are available. Currently, we offer a human library for protein-coding transcripts composed of 16.744 esiRNA and a mouse library for protein-coding transcripts with 14.068 esiRNAs. All esiRNAs are designed based on sequence annotations from ENSEMBL database. The libraries are delivered in 384-well plates with all positions occupied at either 1 µg, 2,5 µg or 5 µg quantities normalized to 50 ng/µl. Other quantities, custom arrays or other plate formats are available upon inquiry.

Specifications:

Quality Control: esiRNAs are designed using the DEQOR design algorithm. The PCR products used for production are tested via gel electrophoresis and identity is confirmed by DNA sequencing. Quality of esiRNA is validated by gel electrophoresis and concentrations are determined by measuring OD260 or fluorescence intensity.
Purification: RNaseIII digested products are purified via column purification using anion-exchange chromatography, and precipitated with isopropanol and finally washed with ethanol.
Format: All esiRNA are delivered in solution (resuspended in nuclease-free TE buffer) normalized to 50 ng/µl concentration.
Storage and stability: esiRNA stored at -20°C have been demonstrated to be stable for 2 years. No decrease in stability after 12 freeze/thaw cycles.
Shipment: Shipped in solution on dry ice in 384-well plates.
Technical Data Sheet: esiRNA will be delivered with an esiRNA Technical Data Sheet, which includes at least plate number and position, esiRNA ID number, organism, amount (µg), concentration (ng/µl), Lot #, and target sequence. Whole Genome esiRNA Libraries will be delivered with a CD containing the product details as given above.
